What Is Enclomiphene?

Enclomiphene is the trans-isomer of clomiphene citrate and belongs to the class of compounds known as selective estrogen receptor modulators (SERMs). It is structurally distinct from zuclomiphene, the cis-isomer, and is primarily studied for its role in influencing the hypothalamic–pituitary–gonadal (HPG) axis.

Unlike mixed-isomer clomiphene, enclomiphene is examined in isolation due to its more targeted receptor behavior and shorter biological persistence.

Mechanism of Action

Enclomiphene functions by antagonizing estrogen receptors at the hypothalamus and pituitary gland. This reduces estrogen-mediated negative feedback, leading to increased secretion of:

  • Luteinizing hormone (LH)

  • Follicle-stimulating hormone (FSH)

The downstream effect under investigation is the stimulation of endogenous testosterone production while maintaining spermatogenesis signaling.

Key Characteristics of Enclomiphene

  • Trans-isomer of clomiphene

  • Classified as a SERM

  • Non-steroidal structure

  • Shorter tissue persistence compared to zuclomiphene

  • Studied for endocrine signaling modulation

Research Applications

Enclomiphene is commonly examined in endocrinology and reproductive biology research, particularly in studies involving:

  • Hypogonadal signaling pathways

  • Estrogen receptor modulation

  • HPG axis regulation

  • Comparative SERM pharmacodynamics

Its isolated isomeric form allows researchers to distinguish effects that may be masked when clomiphene is used as a mixed compound.

Chemical Structure & Properties

Molecular Information

  • Chemical Name: (E)-Clomiphene

  • Molecular Formula: C₂₆H₂₈ClNO

  • Molecular Weight: 405.96 g/mol

  • Compound Class: Triphenylethylene derivative

  • Isomer Type: Trans (enclomiphene)

Structural Notes

Enclomiphene features a triphenylethylene backbone with a chlorine substituent and a tertiary amine side chain. The E-configuration is responsible for its distinct receptor interactions and pharmacokinetic profile compared to the cis-isomer.
